Description: The two forms of pig heart citrate synthase homodimer shown here were determined by crystallographic methods and support the induced fit model of enzyme catalysis. In the absence of CoA-SH the enzyme crystallizes in an “open” form. Citrate binds at the base of large clefts in both catalytic domains of the homodimeric protein. Binding of CoA-SH causes the enzyme to adopt a “closed” conformation, with the clefts essentially filled.
