An especially useful variant of this method is the freeze fracture technique.

If a membrane is frozen quickly and then broken by a sharp blow from a microtome knife, it frequently splits along the plane between the bilayer leaflets

Membranes from individual types of cells or purified organelles can usually be obtained by lysis of the cell or organelle, followed by differential centrifugation.
