Description: When cholesterol levels in the cell are low, Scap transports SREBP to the Golgi. Proteolytic processing by membrane-bound proteases S1P and S2P releases SREBP’s transcription factor domain (bHLH), which enters the nucleus and binds to sterol regulatory elements (SRE) in the promoters of target genes, stimulating their transcription. High levels of cholesterol block this process by Insig-mediated retention of Scap–SREBP in the ER.
