Description: a. Activation. Mediator has three distinct structural domains—head, tail, and middle. The tail domain interacts with transcriptional activators (ACT, in red) and links Mediator with RNA polymerase.

b. Repression. The four-subunit complex containing Srb8, Srb9, Srb10, and Srb11, when bound to Mediator, prevents its interaction with RNA polymerase II and the basal transcription machinery.
