Description: (a) Replication of each fragment terminates with the addition of a ddNTP. Nucleotides of the newly synthesized ″sequenced strand″ are read off the gel. The 5-to-3 polarity of the sequenced strand corresponds to the smaller-to-larger fragment-length direction.
