Answer to Question 1

ANS: C
Nuclear chromatin is initially diffuse or dispersed; as a cell matures, the chromatin undergoes a process of condensing and stains more intensely with Wright stain. The overall cell size decreases; nucleoli are no longer visible if even present, and cytoplasmic basophilia, present because of the high concentration of RNA in a young cell, decrease.

Answer to Question 2

ANS: A
Only the promyelocyte, which is an early granulocyte (neutrophil), can be identified using a Wright stain. None of the stem cells (CFU-E, CSF-GM, HSC) can be identified by morphology. The stem cells are functionally defined by their behavior in vitro using specific colony culture techniques. These require special growth media and specific cytokine (or cytokines). They can be identified using immunophenotypic techniques combined with flow cytometry.
