Answer: Spectral analysis at 450 nm detects methemoglobin.

Answer to Question 2

Answer: The positive sickle solubility test can be confirmed by hemoglobin electrophoresis. The patient's low hemoglobin can affect both results, so packed red blood cells should be used instead of whole blood.
