Insulin is recommended for treatment of type 2 diabetes in patients whose diabetes cannot be controlled by diet or other pharmacotherapeutic agents. If the diabetes can be controlled by diet, the pancreas is still functioning and releasing insulin. Human insulin can be used in gestational diabetes, patients with type 2 diabetes controlled on oral antidiabetic agents with systemic infection, or patients with type 1 diabetes of many years standing.

When depolarization occurs, sodium rushes into the cell. During repolarization, potassium is pumped out of the cell and the resting membrane potential is reestablished. Calcium ions decrease the cell membranes' permeability to sodium and increase the threshold needed to depolarize the cell.
