The CELL-DYN 4000 performs the white count using optical scatter (WOC) and electrical impedance (WIC). Software compares the two as an internal quality control check. When the WOC and the WIC differ, an algorithm selects the most appropriate value for the reported white cell count. Newborn samples, which contain a high concentration of fetal hemoglobin, are an example of when a discrepancy might exist between the two. The soft lysing agent used in the WOC channel is unable to completely lyse the red cells, raising the WOC count compared with the WIC.

When internal instrument checks fail or cast doubt on the data, the automated differential data are suppressed. This means that a manual differential should be performed. Flagged data may be released after appropriate blood film review, as dictated by the specific laboratory policy.
