Answer: Quantification of reticulocytes using manual methods is subject to interobserver variability (during analysis) as well as preanalytical error. Flow cytometry provides a more sensitive and specific means of identifying reticulocytes by identifying the amount of RNA present. It is more accurate because many more cells are counted than in manual methods.
