Answer to Question 1

Acid-fast bacteria has a waxy capsule that is difficult to stain using gram staining but can be stained using concentrated dyes. Once stained, unlike most bacteria, these organisms resist decolorization with the dilute acid and/or ethanol-based solvents (example: Mycobacterium tuberculosis, leprae, and avium complex).
Gram-negative bacteria decolorize by the acetone-ethanol decolorizing solvent and take in the red counterstain, basic fuchsin.
