Water soluble vitamins are absorbed in the GI tract, and then travel through the blood to the cells. Vitamins can travel freely through the blood, and are removed by the kidneys when the levels get too high. Fat-soluble vitamins enter the blood through the lymph, and usually require transport proteins to allow them to travel freely through the blood. Excess fat-soluble vitamins are stored in adipose tissue, and are largely not excreted.
