Answer to Question 1

Answer: The brain is perfused to remove blood from the tissue and is fixed in formalin to harden the tissue and to halt autolysis. Stains are applied to the section to reveal the internal details of the brain section.

Answer to Question 2

Answer: The target of the cannula is decided upon and located in a stereotaxic atlas. The coordinates for placement involve distances in mm from bregma (skull landmark), the midline, and beneath the surface of the skull. A stereotaxic instrument is used to hold the head of an anesthetized rat solidly in place. After incising and retracting the scalp, a hole is drilled through the skull and the cannula inserted into the brain, just above the target site. Dental acrylic and small metal screws are used to hold the cannula in place.
