Drugs that increase receptor activation do not antagonize any natural neurotransmitter in the synapse; this would be an inhibitory action.
Drugs that increase receptor activation do not promote the use of excess transmitter to down-regulate activation.
Drugs that increase receptor activation do not lack any effect at the synaptic cleft.
